"Can a sink with a 2-1/2" drain hole be used with a garbage disposer?"
No, we are not aware of any garbage disposers or adapters that will work with a 2-1/2" drain hole.
If you want to install a garbage disposer, you will need a sink with a 3-1/2" drain hole.
